The specific aims of the proposal are: a) to confirm that LC neurons that exhibit an increase in their firing rate in response to ethanol injections also exhibit significant increases in their excitatory postsynaptic potentials (EPSP) in LC neurons receiving direct inputs from other LC neurons or inputs from other brainstem and spinal cord regions, b) to examine the causal role of the LC in mediating ethanol’s behavioral effects, and c) to examine the action of ethanol within the LC on the release of catecholamines, gamma-aminobutyric acid (GABA), and excitatory amino acids. The proposed studies will be conducted in chronically prepared cat LC-mediating the drug effects, while recording LC neuronal responses using extracellular microelectrodes. Ethanol will be microinjected bilaterally into the LC of lightly anesthetized, paralyzed, artificially ventilated cats. The effect of the systemic administration of drugs and electrical stimulation of LC neurons on ethanol-induced behavioral signs and on the electrophysiology of LC neurons will be determined. In addition to investigating the role of the LC in mediating ethanol’s effects on behavior, this proposal will also address the role of the LC in ethanol’s physiological effects as evidenced by changes in the release of catecholamines, GABA, and the excitatory amino acids glutamate and aspartate. Specifically, the study will investigate the effect of ethanol on the release of these transmitters with the hypothesis that the LC-mediated release of these transmitters plays a critical role in the sedative action of ethanol.
