Extracellular matrix mediates in vitro changes in human corneal endothelial cell sensitivity. To determine whether changes in matrix components that occur in culture influence the sensitivity of human corneal endothelial cells (HCEC) to toxic agents. University-based research laboratory. HCEC were isolated from corneas collected during eye bank procedures and cultured in alpha-modified Eagle medium (alpha-MEM) with bovine serum albumin (BSA) and fibronectin. To test the role of matrix in mediating responses, cells were plated on BSA alone or BSA plus laminin or fibronectin in 96-well plates. After 24 hours, when cells were attached and spreading, media were replaced with serum-free medium containing various concentrations of the toxic agent 5% glucose (heat-killed) or 10% hydroxyethyl methacrylate (HEMA) or HEMA with glutaraldehyde (GA) and the media were added. After 3 days, cell numbers and viability were determined by the clonogenic assay. Preincubation with the matrix components laminin or fibronectin alone significantly reduced the effectiveness of 5% glucose in causing HCEC to decrease in number or become nonviable. However, combined treatment with these components with 5% glucose did not decrease their effectiveness. Preincubation of HCEC with laminin and 5% glucose decreased cell sensitivity to 10% HEMA, with or without GA. In contrast, HEMA alone reduced cell number and decreased viability even in the presence of laminin. The effects of HEMA and GA were additive. However, combining HEMA with fibronectin reduced the effects of HEMA alone. Changes in matrix components alter the sensitivity of HCEC to toxic agents.
